Recombinant tau protein (tau-441 (2N4R) MW 45.9 kDa) was expressed and purified as described77,78. The tau pellet was treated with 8 M urea followed by overnight dialysis against 1X phosphate-buffered saline (PBS) pH 7.4. Tau concentration was measured using bicinchoninic acid protein assay (Pierce BCA Protein Assay Kit, 23225, Thermo Fisher Scientific) and normalized to 1 mg/mL by adding 1X PBS. Aliquots of tau monomer were incubated with heparin (15 kDa) (1:5 molar ratio) at 37 °C on an orbital shaker at a speed of 30 rpm for 5 days as previously described79.
