BM was obtained by flushing donor humerus, tibia, and femur. BM was then RBC lysed and T cell-depleted by labeling cells with biotinylated anti-CD4 and anti-CD8 and anti-biotin microbeads, followed by magnetic cell separation using MS columns (Miltenyi Biotech). In vitro 10 × 106 cells were injected iv into 750 rad lethally irradiated (RS 2000 Biological Research Irradiator), lymphopenic hosts: AbwtTCRα−, AbEpTCRα− and AbEp63KTCRα− mice. Mice were maintained on antibiotic water (1 mg/ml vancomycin) four days prior and one week after transplantation. Mice weight was monitored every 3 days and euthanized when the bodyweight reached 80% of starting weight.
