HMGB1 siRNA transfections

HPMECs were seeded in 6-well plates at a density of 3×10⁵ cells per well in 2 mL antibiotic-free M199 medium with 10% FBS added. HPMECs at 80% confluence were transfected with HMGB1 siRNA transfection reagent according to the manufacturer's instructions. At 24 h post-transfection, LPS and SQV groups were incubated for another 24 h, and then the cells were collected and washed for western blot evaluation. The silencing efficiency was first evaluated with HMGB1 siRNA or HMGB1 siRNA plus 3 μM SQV.
