Primary cultured cerebral cortical neurons (PCNs), obtained from embryonic day 14 ICR mice, were seeded on poly-l-lysine-coated 96-well plates (Sumitomo Bakelite, Tokyo, Japan) at 5 × 104 cells/well or poly-l-lysine-coated 6-well plates (Sumitomo Bakelite) at 1 × 106 cells/well in Neuron medium (Sumitomo Bakelite). Purity of neurons by this method was >98%. PCNs were infected with adenoviruses at the indicated MOI in Neuron medium at 37 °C.
