3,3′-Dihexyloxacarbocyanine iodide (DiOC6(3)) is a lipophilic cationic cyanine dye that is widely used to determine the mitochondrial membrane potential. After treatment and infection, RAW264.7 cells were incubated with DiOC6(3) (40 nM) for 30 min at 37 °C. The cells were then washed with ice-cold PBS and resuspended in 500 μl of PBS. Fluorescence intensities of DiOC6(3) were analyzed on a flow cytometer.
