HepG2.2.15 cells cultured in DMEM supplemented with 10% FBS were seeded in 96-well plates until they reached 40% confluence. IFN-1CTPON, IFN-2CTPON, or rhIFN-α2b (as the standard) serially diluted 40–49-fold from 400 ng/ml was then added and the cells incubated for 72 h, with four parallel wells for each IFN dilution. The HBsAg concentration in the supernatant from each well was measured with a commercially available ELISA kit (Wantai Pharmaceutical Co., China).
