CCK-8 assay

Cell proliferation abilities of the MCF-7 or MDA-MB-231 cells were evaluated by Cell Counting Kit-8 assays. Cells were transfected as mentioned above. Post-transfection at 48 h, the cells were digested, resuspended into single-cell suspension, seeded into 96-well plates with 100 µl of cells per well (2×10⁴ cell/ml), and incubated at 37°C with 5% CO₂. CCK-8 reagent (10 µl) was added to per well after incubation at 0, 24, 48, 72 and 96 h, and incubated for 1 h for each time point. Absorbance was measured via a microplate reader (Bio-Rad Laboratories) at 450 nm.
