N. benthamiana wild-type and transgenic line Nb308A plants were used in agroinfiltration experiments as described previously (Li et al., 2014). Briefly, A. tumefaciens cells were firstly inoculated into LB medium to grow overnight; the cultured bacterial cells were harvested and diluted in fresh LB medium to OD600 = 0.1 and grown for additional 6–8 h. The bacteria were then resuspended in H2O and infiltrated into the underside of fully expended N. benthamiana leaves using a syringe. The infiltrated plants were placed at 22°C in a 16-h light/8-h dark photoperiod.
