The number of colony-forming units (CFUs) was evaluated for MSCs from whole, concentrated and clotted V-BMAs. Two-hundred MSCs/cm2 were plated onto six-well plates and cultured for 10 days. At the endpoint, cells were fixed with 10% formalin for 20 min and stained with 0.1% toluidine blue in 1% paraformaldehyde (PFA) for 1 h12,14. The aggregates with ≥20 cells were visually scored as colonies and counted with a light microscope (Olympus BX51).
