The Yeast Tet-promoters Hughes Collection (yTHC) was purchased from GE Healthcare Dharmacon. A total of 800 essential genes are included in the collection. For each essential gene, a kanR-tetO7-TATA cassette was used to replace the endogenous promoter of the gene in the haploid strain R1158 (URA3::CMV-tTA MATa his3-1 leu2-0 met15-0). The diploid yeast strain used in this study is BY4743 (MATa/α his3Δ1/ his3Δ1 leu2Δ0/ leu2Δ0 lys2Δ0/ lys2Δ0 metΔ15/met15 ura3Δ0 /ura3Δ0).
