The in vitro angiogenic potential of EPCs was performed by matrigel tube formation assay. Briefly, 24-well plates were coated with matrigel (250 µL/per well, BD Biosciences, USA). After transduced, EPCs were digested and adjusted to a concentration of 2×105 cell/mL, consequently 5×104 cell/well was seed, following incubating at 37 °C with 5% CO2 or hypoxia for 6 h to form tubes. Capillary-like structures were imaged and counted.
