Total DNA was extracted from two-copy and four-copy mouse adrenal glands using DNeasy tissue kit following manufacturer’s instructions (Qiagen, Waltham, Massachusetts, USA). qPCR analysis was done using 2.5 ng of total DNA as template and primers specific for mitochondrial (MtND1) and nuclear (Gapdh) genes. Relative amount of mitochondrial DNA to that of nuclear DNA was calculated by 2−ΔΔCt method and changes were expressed in arbitrary units (AU).
