4.7. Proliferation Assay

Mouse 4T1-shCTR and 4T1-shStat3 tumor cells were seeded at a density of 4 × 10³ cells per well containing 100 uL in 96-well plates with complete RPMI-1640 medium for three days. For each cell line and each time point (0, 24, 48 and 72 h) six replicates were used. A mixture of XTT labeling with electron-coupling reagents was then made according to manufacturer’s protocol (Cell Proliferation Kit II (XTT), Roche, Basel, Switzerland). For each time point, measurement of cell viability was performed via microplate reader at a wavelength of 490 nm (reference wavelength: 650 nm).
