5 million tumor cells in inoculation media (HBSS+Matrigel 1:1 mix) were implanted subcutaneously in the right hind flank of female NIH nude rats (Taconic Biosciences). When tumors reached ∼400 mm3, animals were randomized into groups of 8–10 and treated as indicated in the respective figure legends. LY3009120 was administered orally and animals were monitored for toxicity as previously described [15, 53]. Vehicle control was 20% Captisol® for studies with Colo 205, HT-29 and HCT 116 and 1% HEC/0.25% Tween 80/0.05% Antifoam for the Colo 320HSR study. All procedures and techniques were performed in accordance with the institutional guidelines of the American Association for Laboratory Animal Care and monitored by the Eli Lilly and Company Animal Care and Use Committee.
