The focal cerebral ischemic model of rat was induced by MCAO as described previously42. Briefly, the rats were anesthetized with 2% chloral hydrate. The right middle cerebral artery of the rats was occluded using a 3-0 nylon monofilament suture (Ethicon Nylon Suture, Ethicon Inc., Sukagawa, Japan). Regional cerebral blood flow (rCBF) was monitored using a disposable microtip fiberoptic probe (diameter, 0.5 mm) connected through a master probe to a computerized laser Doppler main unit (PeriFlux 5000, Perimed AB, Sweden). Rats retaining >20% of baseline perfusion during ischemia were excluded. The temporalis muscle temperature was monitored and was maintained at 37.0 °C–37.5 °C by surface heating or cooling during surgery. After 120 min of ischemia, reperfusion was done. At 120 min after reperfusion, the rats received EA treatment. After that, rats were then allowed to recover before being returned to their home cages.
