Semi-quantitative RT-PCR analysis of alternative splicing

RT-PCR analysis was performed using amplimers based on the sequences of Adam15 exons 19 and 21 (svADAM15F1: 5’-GCGGGCACAGCAGATGAC-3’ and svADAM15R1: 5’- GGGTTGGCAGGCAGTGGC-3’) and Yap1 exons 5 and 7 (svYap1F1: 5-GGAGAAGGAGAGACTG-3’ and svYap1R2: 5’-GTCCCTCCATCCTGCTC-3’). The PCR products were examined by agarose gel electrophoresis and staining with ethidium bromide. Adam15 mRNA and exon 20-skipped Adam15 mRNA were detected as 277 bp and 78 bp DNA fragments. Yap1 mRNA and exon 6-skipped Yap1 mRNA were detected as 144 bp and 96 bp DNA fragments.
