[3H](+)-Pentazocine binding assays

For competition binding assays, the radioligand used was [3H](+)-pentazocine, with a specific activity of 32.2 Ci/mmol (PerkinElmer Life Sciences, Boston, MA, USA). Experiments were performed in crude synaptosomal membranes (P2 fraction) from wild-type mouse brain, according to a previously described method14,16,21. Detailed methods are reported in the Supplementary Information.
