Parental HCC1806 and HCC1599 were used to generate cells with doxycycline-inducible knock-down of MLK4. HEK293T cells were transfected with human TRIPZ shRNA against MLK4 (Dharmacon) to generate a lentiviral stock. The sequences of shRNA are as follows: sh2_ATCAGAATGTTAAGTTCCC and sh6_TCTTGATACACTACAATCA. Cells were transduced with lentiviral stocks and cell lines generated by antibiotic selection with puromycin. Doxycycline (Sigma) in 1 µg/ml concentration was used to induce knock-down of MLK4.
