As described previously (8), kinetic solubility was measured by diluting a 10 mM dimethyl sulfoxide (DMSO) stock solution of the test compound down to 100 μM in PBS (pH 7.0) or 0.1 N HCl (pH 1.0). The samples were incubated at 37°C for 1 h, followed by centrifugation at 2,800 relative centrifugal force (RCF) for 30 min at room temperature. Thermodynamic solubility was determined with the crystalline-free base of GS-6620 or amorphous GS2 using the shaking flask method. The concentrations of the compounds in supernatants were determined by HPLC.
