Microglial BV-2 cells were plated in 12-well plates (1 × 105 cells/well) and were transiently transfected with siRNA for 30 min, using a mixture of CHI3L1 siRNA (20 nM) and the RNAiMAX reagent in OPTI-MEM, according to the manufacturer’s specification (Invitrogen). The transfected cells were treated with K284-6111 (0.5 μM) for 24 h and then used for detecting protein expression.
