The effect of Piperine on the viability of cells was detected by MTT assay. Briefly, A2780 cells and OSE cells at the logarithmic phase were seeded in a 96-well plate at a concentration of 1 × 104/well in a 96-well plate overnight. After being treated with Piperine for 12, 24, 48, and 72 h at the concentration of 20 μM, or with indicated concentrations of Piperine (0, 4, 8, 16, or 20 μM) for 48 h, 20 μl of MTT solution (5 mg/ml in PBS) was added to each well and incubated for another 4 h at 37°C. Then, the MTT solution was removed and DMSO (100 μl) was added to each well to dissolve the MTT formazan. The absorbance was measured at 540 nm with a microplate reader.
