Transfection of cells with siRNA was performed using Lipofectamine® 2000 transfection reagent as described previously [19, 21]. Commercially available human MerTK and negative control siRNA were used for transfection at indicated concentrations. Briefly, at 16 h after transfection, cells were treated with sulforaphane for 30 min prior to treatment with Aβ1-42 for 16 h. Levels of IL-1β or TNF-α in culture supernatant were analyzed using human-specific IL-1β or TNF-α ELISA kit (BD Biosciences).
