Nuclear extract of Arabidopsis plants (7 d old) was prepared using the Epiquick Nuclear Extraction Kit I (Epigentek), and total histone deacetylase activity in nuclear extracts was measured in the presence or absence of recombinant 32E03 (500 or 1500 nM) or trichostatin (500 nM) using the Epigenase HDAC Activity/Inhibition Direct Assay Kit (Epigentek). Protein concentration in the nuclear extract was determined using the Coomassie Protein Assay Reagent (Thermo Fisher Scientific).
