The survival of the cells grown on the 3D scaffolds was assessed using the Live/Dead Cell Staining Kit. The cell-carrying scaffolds at different time points or under different treatment conditions were collected and manually sectioned at a thickness of 250 to 500 μm. The sections were washed twice with 1× PBS (37°C), incubated in the staining solution (2 μM calcein-AM and 4 μM EthD-III in 1× PBS) at RT for 45 min, and imaged using fluorescence microscopy. Calcein-AM stains live cells green (under EGFP filter), whereas EthD-III stains dead cells red (under Texas Red filter).
