Three hundred microliters of HRMVEC suspension at a concentration of 5 × 105 cells/ml were added to each well of a twenty four-well culture plate that was pre-coated with 280 μl of growth factor-reduced Matrigel. Cell were treated with vehicle or VEGFA (40 ng/ml) for 6 hrs at 37 °C. Tube formation was observed under an inverted microscope (Eclipse TS100, Nikon, Tokyo, Japan) and the images were captured using a CCD color camera (KP-D20AU: Hitachi, Ibaraki, Japan) and Apple iMovie 7.1.4 software. The tube length was calculated using NIH ImageJ and expressed in micrometers.
