To create resistant lines, parental cells were cultured with increasing concentrations of osimertinib, starting at the IC30 (19). At 80-90% confluence, cells were trypsinized and divided into 2 tubes. Half were frozen and the rest were reseeded into a new dish at a 20-30% higher dose of osimertinib. Fresh drug was added every 72 – 96 hours.
