Male C57Bl/6J mice (Jackson Laboratories, Bar Harbor, ME) or MaFIA (Macrophage associated Fas-Induced Apoptosis, #5070Jackson Laboratories, Bar Harbor, ME) were started on either a low fat diet (LFD; 10% Kcal, Research Diets #12450D) or high fat diet (HFD; 60% Kcal, Research Diets #D12492) beginning at 4-weeks of age, and were maintained on their respective diets for the duration of the experiment. Male C57Bl/6J mice were used as this strain and sex are very susceptible to diet-induced obesity and T2DM [8, 9]. MaFIA mice express Green Fluorescent Protein (GFP) under the control of the Csf1r-promoter resulting in GFP expression in macrophages and dendritic cells. Mice were maintained in pathogen free housing on 12hr/12hr light/ dark cycle with ad libitum access to food and water (except during fasting when food was removed).
