Cell culture

Human OS cell lines SaOS (p53-null), 143B, HOS, U2OS (wild-type p53), MG-63 (mutant p53) and human osteoblasts (CRL-1132) were purchased from ATCC (Manassas, VA). SaOS and U2OS cells were cultured in McCoy’s 5A medium, 143B, HOS and MG-63 cells in Modified Eagle’s Medium (MEM) and osteoblasts in Ham’s F12 Medium. Growth media were supplemented with 10% fetal bovine serum (FBS), 2 mM L-glutamine, 25 U/mL penicillin, and 25 μg/mL streptomycin. Cells were maintained at 37°C in humidified incubators, in an atmosphere of 5% CO₂.
