The cytotoxicity of glecaprevir for Huh-7, HepG2, and MT4 cells was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT; Sigma-Aldrich, St. Louis, MO) colorimetric assay. Huh-7 cells containing the genotype 1a replicon or HepG2 cells (4,000 cells/well) were plated in 96-well plates and incubated overnight before the addition of glecaprevir. For MT4 cells, 10,000 cells/well were plated immediately before the addition of glecaprevir. The rest of the assay was performed as described previously (33).
