TsVSVG fused to GFP, pEGFPN1-VSVG, was purchased from Addgene (11912; Cambridge, MA) and has been described previously (Presley et al., 1997 
). MDA-MB-231 or paxillin–/– MEFs plated on glass coverslips were infected with the adenovirus overnight in a 40°C incubator. Cells were subsequently transferred to 32°C to release VSVG from the ER for various time periods, as indicated, before fixation and immunofluorescence microscopy or were subjected to live-cell imaging.
