Flies with genotype UAS-pros.S; act5C-GAL80ts-insulated spacer-tub-GAL80ts/dpn-LexA::P65; 13XlexAop-Syp-miRNA/dpnEE-Gal4 were cultured at 18°C. The GAL80 and non-temperature-sensitive LexA::p65 allowed Syp knockdown continuously. For the control experiment, collected white pupae were cultured at 18°C for 24 h before dissection. To induce ectopic Pros in NBs, collected white pupae were transferred to 29°C to inactivate the temperature sensitive GAL80 for 16 h before dissection and thus de-repress dpnEE-Gal4 for misexpression of Pros.
