The cell lines were trypsinized and 3 × 104 cells were suspended in DMEM containing 0.4% low melting point (LMP) agarose (Promega, Madison, WI, USA). Two milliliter of the above cell suspension was plated over 3 mL bottom agar containing 0.8% LMP agarose in a 60‐mm dish. Two independent experiments were performed in quadruplicate. The dishes were incubated at 37 °C with medium replenished every two days until colony formed. The colonies were stained with 0.01% crystal violet solution. Representative pictures were taken under stereomicroscope (Olympus SZX10, Center Valley, PA, USA).
