140,000 cells in 1mL of complete medium were plated into each well of 12 plastic plates (Grenier Bio-one, Cat-No# 665180) and grown for 24 hours. Lipofectamine LTX (Invitrogen, Cat-No# 15338-100) was used for transient transfection according to the manufacturer’s protocol. In short, the transfection mix was prepared by adding 200ng of plasmids in 200µL of DMEM for each well. 1.5µL of Lipofectamine LTX was added to the mix and incubated at room temperature for 30mins. The transfection mix was then applied to the plates and mixed by gentle shaking. Dox induction was performed after 2 hours. The cells were incubated for about 48 hours before analysis.
