Isolated pancreatic islets were homogenized in a RIPA buffer containing phosphatase and protease inhibitors. Homogenates were subjected to SDS-PAGE (10–12 %), transferred to a PVDF membrane and immunoblotted with polyclonal α-CST (CgA352–372) (Pasqua et al. 2016) and α-C-terminal (CT)-CST (CgA368–373) (Bianco et al. 2016) and monoclonal α-VS1 (CgA47–57) antibodies (Colombo et al. 2002; Ratti et al. 2000).
