In total, 40 µM of 13C-formaldehyde was added to complete IMDM, including 10% FBS, and 1 ml of this medium was added to blank wells of a 12-well plate or to wells in which 100.000 HAP1-wt cells were seeded the day before the experiment. Plates were cultured in an incubator at 37 °C in a humidified 5% CO2 atmosphere. At the indicated time points, the medium was harvested, cells spun down and the supernatant stored at −80 °C, before 100 µM of this were used for the formaldehyde quantification. The experiment was performed three times, with three replicates per experiment.
