For flow cytometry analysis, at least three leaves per time point of a biological repeat (n = 3) were chopped with a razor blade in 200 µL of Cystain UV Precise P Nuclei Extraction buffer (Partec), followed by the addition of 800 µL of staining buffer and filtering through a 50-µm filter. Nuclei were analyzed with the Cyflow MB flow cytometer (Partec) and the FloMax software, and the statistical analysis (ANOVA, Dunnett’s test) was performed in GraphPad Prism.
