Using the cell models described above, we assessed Rap1 activation by the Rap1 activity assay with a kit obtained from Millipore according to manufacturer instructions. Rap1-GTP and total Rap1 protein content was determined by Western blot analysis. ChemiDoc imaging system (Bio-Rad) was used to image gels and quantification of protein expression was accomplished by densitometry software (Quantity One; Bio-Rad).
