Mice were randomized to either miR-182 antagomir or MM control treatment, delivered into brains via stereotactic ICV injection as described previously [19]. Briefly, mice were anesthetized and positioned in the stereotaxic injection device. The skull was exposed with a surgical incision and a burr hole was made in the left side at the following stereotaxic coordinates: bregma: − 0.58 mm; dorsoventral: 2.1 mm; lateral: 1.2 mm [20]. MiR-182 antagomir (3 pmol/gram in 2 μl final volume) or mismatch control (same amount) was mixed with the transfection reagent DOTAP (4 μl; Roche Applied Science, San Francisco, CA). The total volume of the mixture (6 μl) was infused into the left ventricle over 20 min. The skull was then sealed with bone wax and the incision sutured, and the animal was allowed to recover for 24 h prior to MCAO.
