Hearts were loaded with the fluorescent intracellular Ca2+ ([Ca]i) indicator Rhod-2AM and the voltage-sensitive dye RH237. Blebbistatin (10µmol/L) was used to eliminate motion artefacts during wide-field optical imaging of epicardial transmembrane potential (Vm) and [Ca]i using a dual Micam-Ultima complementary metal–oxide semiconductor camera system on a THT macroscope (SciMedia, Costa Mesa, CA).
