Sulfotransferase activity toward chondroitin was assessed using an established method62 with slight modifications. Briefly, the reaction mixture contained 10 µl of resuspended beads, 50 mM imidazole-HCl (pH 6.8), 2 mM dithiothreitol, 10 µM [35S]PAPS (3 × 105 dpm), and chondroitin (1 nmol as GlcA). The reaction mixtures were incubated at 37 °C for 30 min and gel filtered using a disposable syringe column. Radioactivity in the flow-through fractions was measured by liquid scintillation counting.
