We obtained the Trastuzumab binders’ dataset (11,300 sequences) from the Github repository (https://github.com/dahjan/DMS_opt) from Mason et al. (47) For plotting the binders sequence logo in Figure 3B , we converted the sequences to a position specific scoring matrix. For plotting the DMS logo in Figure 3B , we obtained the single-site DMS data directly from the authors.
