Agonist-induced platelet aggregation according to Born [20] was measured as described previously with some modifications [21]. The platelet count in platelet-rich plasma was adjusted to 2 × 108/mL with autologous platelet-poor plasma to compare platelet aggregation responses with lumi-aggregometry analysis (Section 2.10). ADP (5 µM, Sigma-Aldrich, St. Louis, MO, USA), the thrombin receptor PAR-1-activating peptide TRAP-6 (20 µM, Bachem Biochemica GmbH, Heidelberg, Germany), epinephrine (2 µM, Sigma-Aldrich), and the thromboxane A2 (TxA2) mimetic U46619 (3 µM, Sigma-Aldrich) were used as platelet agonists according to the recommendation of the SSC “Platelet Physiology” of the ISTH [22].
