All SegModTeX reactions were conducted according to the following optimized protocol: 1 x ThermoPol buffer (NEB), 15 mM MgSO4, 1.5 mM rNTPs, 0.1 mM RNA seg:ssDNA template, and TGK polymerase to a final concentration of ~0.1 mg/ml were mixed at 4 °C. For large RNAs, SUPERase•In™ RNase Inhibitor was added according to the manufacturer’s specifications.
