HEK293T cell culture

HEK293T cells were purchased from ATCC (CRL-3216) and validated by Short Tandem Repeat (STR) profiling. Mycoplasma testing was negative. Wild-type or engineered HEK293T cell lines were maintained at 37°C, 5% CO₂ in Dulbecco’s modified Eagle medium supplemented with 10% fetal bovine serum, 1% GlutaMax-I, penicillin (100 U ml⁻¹), and streptomycin (100 μg ml⁻¹). For maintaining or expanding the cell culture, we used TC-treated culture dishes (Corning). For all imaging experiments, cells were plated on PDL-coated glass-bottomed dishes (Cellvis, Cat.# D35-14-1.5-N).
