The pLOU3-waaB plasmid was used as the template for mutagenesis which was performed based on the protocol of Liu and Naismith (2008), a modified version of QuikChange Site-Directed Mutagenesis System. All mutations were confirmed by DNA sequencing. The mutant plasmids were transformed into the BL21 (DE3). The expression and purification of WaaB mutant proteins were performed using the same protocol as that of the wild-type WaaB.
