In situ hybridization

RNAscope in situ hybridizations for freshly frozen tissue were performed according to the manufacturer instructions (ACDBio; Table 1). The Piezo2 probe was tested in TG slices of adult wild-type littermates and Piezo2^cKOmice. Slices of 20 µm were cryosectioned in O.C.T. and stored at −80°C. Sections were thawed just before staining and fixed with 4% PFA for 15 min followed by rinsing in PBS. All sections were mounted with Fluoromount Mounting Medium (Sigma-Aldrich).
