To assess actual cleavage sites in neuropeptide precursors in an unbiased way, we focused on neuropeptide structures that had been identified by mass spectrometry-based systematic peptide identification. AA sequences of neuropeptides were acquired from Homo sapiens, Drosophila melanogaster (Arthropoda) and Lineus longissimus (Nemertea). N-terminal and C-terminal cleavage sites (2 AA) and flanking regions (6 AA) of the collected neuropeptides in their precursors were extracted and subjected to composition analysis or sequence logo analysis with WebLogo57. The sources of peptide information for each animal species and the details of the cleavage site analysis are described in the Supplementary Information.
