Treg migration was assayed using 24-well Transwell chambers with 5 μm pores. A total of 105 cells in 100 μL of media (RPMI 1640 with 0.5% BSA) were placed in the upper chambers. Aggregated α-syn, diluted in 600 μL media, was placed in the lower wells, and the chambers were incubated at 37 °C. After 3 and 4 h, the migrated cells present in the bottom wells were counted using a LUNA-II automated cell counter (Logos Biosystems, Anyang, Republic of Korea).
