Peripheral blood mononuclear cells (PBMCs) (10 ml) were isolated using Ficoll-Hypaque solution (Haoyang biotech, Tianjing, China), and cultured in RPMI-1640 at 37 °C in 5% CO2 for 2 h until the monocytes adhered to the bottom of culture flasks. Then the lymphocytes suspended in middle were isolated. The purity was more than 90% through flow cytometry assay and the viability was more than 90% through Trypan blue staining. Cells were cultured at 37 °C in RPMI 1640 medium, supplemented with penicillin-streptomycin prior to the analysis.
