S2 cell culture, transfection and dsRNA treatment

S2 cells were cultured in Schneider’s Buffer, 10% FBS, 1% Penicillin and Streptomycin, 50 µM Tetracyclin at 25°C. Qiagen Effectene and was used as a transfection reagent according to the producer’s instruction. Mfn DsRNA were designed according to a protocol from www.flyrrnai.org and synthesized using the Mega T7 kit from Ambion. Cells were harvested 96 hr after dsRNA treatment.
